City fire spokesman Erik Scott says the blaze broke out around 3 a.m. Thursday at the two-story hotel in the Wilmington area south of downtown.
It took more than 100 firefighters about an hour to extinguish the flames.
Scott says a man died at the scene. Two other people were seriously hurt, and nine others received minor injuries. Scott says a child who's about 8 years old is among the injured.
More than 15 people have been displaced from the 20-unit Wilmington Hotel just north of the Port of Los Angeles.
The cause is under investigation. Scott says the hotel had functional smoke alarms.
